The macrophage checkpoint SIRPα pathway is a critical immune regulatory axis involving the interaction between Signal Regulatory Protein Alpha (SIRPα) and its ligand CD47. This pathway functions as an inhibitory "don't eat me" signal that prevents macrophages and other myeloid cells from phagocytosing healthy self-cells, but is also exploited by cancer cells to evade immune clearance.
