Overview

Macrophage colony-stimulating factor 1 receptor (CSF1R), also known as M-CSFR, CD115, or c-FMS, is a cell-surface transmembrane protein encoded by the CSF1R gene. It is a member of the type III protein tyrosine kinase receptor family and plays a central role in regulating the survival, proliferation, and differentiation of myeloid lineage cells, especially macrophages and microglia. Upon ligand binding (CSF-1 or IL34), the receptor dimerizes and activates downstream signaling pathways including PI3K/Akt, JNK, ERK/MAPK pathways. Mutations in CSF1R are linked to several diseases including ALSP, cancer, and skeletal disorders. Therapeutic targeting of CSF1R is under clinical investigation for various diseases.
