CSF1R is a type III transmembrane protein tyrosine kinase receptor primarily expressed on myeloid lineage cells. Ligand binding (CSF-1 or IL-34) leads to receptor dimerization, autophosphorylation, and activation of downstream signaling pathways (PI3K/Akt, JNK, ERK1/2) regulating cell survival, proliferation, differentiation, migration and function of myeloid cells. It is implicated in diseases like ALSP, leukodystrophy, skeletal dysplasia and cancer.
