Macrophage polarization is a dynamic process in which macrophages adopt distinct functional phenotypes (M1/M2) in response to microenvironmental cues. M1 macrophages are pro-inflammatory and microbicidal, while M2 macrophages are anti-inflammatory and involved in tissue repair. This pathway is regulated by various ligands (LPS, IFN-γ, IL-4, IL-10) binding to their respective receptors (TLRs, IFNGR, IL4R, IL10R) and activating downstream signaling pathways (NF-κB, STATs, PI3K/Akt/mTOR, PPARs). Dysregulation of macrophage polarization contributes to various diseases, making it a promising therapeutic target.
