Macrophage metalloelastase (MMP-12) is a zinc-dependent endopeptidase belonging to the matrix metalloproteinase family. Primarily expressed by mature tissue macrophages, it exhibits significant elastolytic activity, degrading elastin and other ECM components. MMP-12 plays a crucial role in tissue remodeling during inflammation and injury, facilitating macrophage migration. It is implicated in various diseases, including emphysema, atherosclerosis, and rheumatoid arthritis. Expression is modulated by factors like hyaluronic acid fragments, interferon-gamma, and dexamethasone. Loss-of-function studies highlight its importance in macrophage-mediated ECM proteolysis and tissue invasion.
