Macrophage migration inhibitory factor (MIF) mRNA is the transcript encoding a pleiotropic pro-inflammatory cytokine and enzyme that plays a central role in the regulation of innate and adaptive immunity. MIF is unique among cytokines for its ability to counter-regulate the anti-inflammatory effects of glucocorticoids, effectively acting as a pro-inflammatory primer in various disease states. The mRNA itself serves as a therapeutic target for gene-silencing technologies, such as small interfering RNA (siRNA) and antisense oligonucleotides (ASOs), which aim to suppress the de novo synthesis of the MIF protein. Elevated levels of MIF mRNA and its translated protein are strongly associated with the pathogenesis of chronic inflammatory conditions like rheumatoid arthritis and sepsis, as well as multiple cancers where MIF promotes tumor growth, angiogenesis, and immune evasion. While most clinical-stage interventions currently focus on neutralizing the MIF protein or blocking its primary receptor, CD74, mRNA-targeted approaches offer a promising strategy for achieving potent and sustained inhibition of the MIF signaling axis in refractory diseases.
Drugs targeting MIF mRNA primarily utilize RNA interference (RNAi) or antisense mechanisms to induce the degradation of the transcript or block its translation, thereby reducing the production of the pro-inflammatory MIF protein. siRNA-based therapies recruit the RNA-induced silencing complex (RISC) to cleave the mRNA, while antisense oligonucleotides (ASOs) can trigger RNase H-mediated degradation or provide steric hindrance to the ribosome.
