Overview

A "macrophage stimulant" refers to any molecule, cytokine, or agent that activates or enhances the function of macrophages—key immune cells responsible for engulfing pathogens, clearing debris, and orchestrating immune responses. The most well-characterized endogenous stimulants are colony-stimulating factors (CSFs), particularly Macrophage Colony-Stimulating Factor (M-CSF/CSF1) and Granulocyte-Macrophage Colony-Stimulating Factor (GM-CSF/CSF2). M-CSF primarily promotes macrophage differentiation, survival, and M2 polarization, while GM-CSF drives pro-inflammatory M1 activation. Agents targeting the CSF1/CSF1R axis are under investigation for cancer therapy and other immune-related diseases.
