Overview

MAGE family member D4 (MAGE-D4) is a cancer-testis antigen and a member of the MAGE (melanoma antigen gene) superfamily, which comprises proteins with restricted normal tissue expression (primarily testicular/placental) but frequent reactivation in various cancers[2][3][4][5]. MAGE-D4 is a nuclear protein involved in the negative regulation of transcription[3]. As with other MAGE proteins, it may act as a regulator of E3 ubiquitin ligase activity, modulating the stability of target proteins and influencing processes such as cell proliferation, differentiation, and apoptosis[2]. MAGE-D4 is aberrantly expressed in several tumor types and has been identified as a marker of poor prognosis and a potential therapeutic target, especially in solid tumors such as breast cancer[4]. Like other MAGE antigens, MAGE-D4 is under investigation as a diagnostic/prognostic biomarker and potential target for immunotherapy, reflecting its value as a cancer-testis antigen.
