MAGE family member E2 (MAGEE2) is a protein encoded by the intronless MAGEE2 gene, part of the E-type subfamily of the MAGE (melanoma antigen-encoding gene) family. The MAGE proteins are characterized by the MAGE homology domain and interact with E3 ubiquitin ligases, influencing protein stability and cell signaling, with important roles in tumorigenesis. MAGEE2 has been associated with melanoma and hepatocellular carcinoma and may play roles in transcriptional processes and brain development, particularly in rodents. While extensively studied in the context of cancer-testis antigens, specific therapeutic targeting and unique mechanistic details for MAGEE2 remain less well-defined relative to other MAGE family members[5][7][1][2][3][4].
As a member of the MAGE family, may act by influencing the activity of E3 ubiquitin ligases, impacting protein ubiquitination and degradation in cancer cells (e.g., impacting p53, as seen in other MAGEs)[1]
