MAGEA4 antisense RNA 1 (MAGEA4-AS1) is a long non-coding RNA transcript of approximately 418 nucleotides, found predominantly in the cell nucleus. It is upregulated in oral squamous cell carcinoma (OSCC) tissues compared to normal controls, and its increased expression is associated with enhanced proliferation, migration, and invasion of OSCC cells. Mechanistically, MAGEA4-AS1 interacts with the tumor suppressor protein p53 and enhances p53’s activation of the MK2 gene, thereby promoting downstream signaling pathways that facilitate tumor growth and metastasis. Functional studies indicate that knockdown of MAGEA4-AS1 diminishes these tumor cell behaviors, supporting its evaluation as both a biomarker for OSCC and a candidate therapeutic target.
Molecular mechanism involves binding to p53, which in turn activates MK2 transcription, promoting tumor proliferation and metastasis.
