Overview

MAGI2 antisense RNA 3 (MAGI2-AS3) is a long non-coding RNA (lncRNA) transcribed from the 7q21.11 chromosomal locus and is involved in the post-transcriptional regulation of gene expression, primarily by functioning as a molecular sponge for multiple microRNAs[1][2][3][5]. This lncRNA is found in both the nucleus and cytoplasm, depending on the cell type[2]. MAGI2-AS3 is aberrantly expressed in various cancers, where it typically acts as a tumor suppressor, inhibiting cell proliferation, migration, invasion, and promoting apoptosis via interactions with cancer-associated microRNAs and regulation of downstream mRNAs[1][2][3][4][5]. Expression of MAGI2-AS3 is also linked to non-malignant conditions, including congenital diaphragmatic hernia, Alzheimer’s disease, and intervertebral disc degeneration[1]. Its expression can be epigenetically silenced by DNA methylation, which can be pharmacologically reversed[2]. Mechanism of action
