Target intelligence / Profile preview
Magnesium ion homeostasis is the physiological process responsible for maintaining stable magnesium concentrations in the blood and tissues, which is vital for numerous cellular functions. This pathway is regulated through a balance of intestinal absorption, renal excretion, and mobilization from bone stores, primarily mediated by specific ion channels and transporters such as TRPM6, TRPM7, and SLC41A1 (de Baaij et al., 2015). Magnesium acts as an essential cofactor for over 300 enzymes, including those involved in ATP metabolism, DNA and RNA synthesis, and protein synthesis (NIH Office of Dietary Supplements). Disruptions in this pathway can lead to hypomagnesemia or hypermagnesemia, contributing to conditions like cardiovascular disease, type 2 diabetes, and neurological disorders (Romani, 2011). Pharmacological management typically involves magnesium supplementation or the adjustment of medications that impair magnesium handling, such as diuretics or proton pump inhibitors (William & Danziger, 2014). Understanding this pathway is crucial for managing electrolyte imbalances and their systemic consequences.
Drugs affecting this pathway act by supplementing systemic magnesium levels, inhibiting renal reabsorption in the thick ascending limb of Henle, or disrupting active transport in the gastrointestinal tract.
