Magnetic Resonance Imaging (MRI) signal is the radiofrequency energy emitted by atomic nuclei, typically hydrogen protons, as they return to an equilibrium state after excitation by radiofrequency pulses within a powerful magnetic field (NIBIB, 2023). This signal is not a biological therapeutic target such as a receptor or enzyme; rather, it is a physical phenomenon used to non-invasively visualize internal body structures and physiological processes (StatPearls, 2023). In clinical and research settings, the signal's characteristics—such as T1 and T2 relaxation times—are exploited to differentiate between healthy and diseased tissues (Mayo Clinic, 2023). Pharmacological intervention involves the use of contrast agents, which do not bind to the signal as a 'target' but instead modify the local magnetic environment to enhance signal intensity and diagnostic clarity (PubChem, 2024). Consequently, while the MRI signal is indispensable for monitoring disease progression and drug efficacy, it represents an imaging modality output rather than a target for therapeutic drug design.
Contrast agents interact with the magnetic environment of water protons to shorten longitudinal (T1) and transverse (T2) relaxation times, thereby increasing or decreasing the MRI signal intensity to improve image contrast.
