Overview

The Human papillomavirus type 51 L1 protein is the primary structural component of the HPV 51 virion, a high-risk genotype associated with cervical cancer. This protein self-assembles into pentameric capsomeres, which further organize into an icosahedral capsid to encapsulate the viral genome. The L1 protein plays a critical role in the viral life cycle by mediating the initial binding to heparan sulfate proteoglycans on the basement membrane of the host epithelium, facilitating subsequent infection. [1][2] In a clinical context, the L1 protein is a vital target for prophylactic vaccines. While current widely available vaccines like Gardasil 9 do not include HPV 51, next-generation multivalent vaccines, such as the 14-valent (SCT1000) and 11-valent (RECO-201) candidates, specifically incorporate HPV 51 L1 virus-like particles (VLPs). These VLPs are non-infectious but highly immunogenic, stimulating the production of neutralizing antibodies that block the virus from entering host cells, thereby preventing infection and the subsequent development of associated malignancies. [3][4]
