Overview

Major facilitator superfamily domain-containing protein 10 (MFSD10) is a member of the major facilitator superfamily of transporters. It is primarily expressed in the kidney, localizing to the luminal membrane of proximal tubules, where it functions as a transporter for organic anions, including the non-steroidal anti-inflammatory drugs indomethacin and diclofenac. MFSD10 is capable of both efflux and uptake, thus influencing the renal excretion and systemic pharmacokinetics of substrate drugs. Overexpression of MFSD10 confers cellular resistance to specific NSAIDs, indicating an efflux pump function akin to its yeast orthologue TPO1. The transporter’s activity may impact drug bioavailability and side effects, and its molecular structure supports its role as part of the major facilitator superfamily.
