Major facilitator superfamily domain-containing protein 12 (MFSD12) is a transmembrane cysteine transporter protein expressed in lysosomes and melanosomes, key for regulating melanin biosynthesis and maintaining lysosomal cysteine homeostasis. MFSD12 is essential in pigment cells for the import of cysteine, which modulates skin pigmentation via the synthesis of pheomelanin, and is also critical in non-pigmented cells for maintaining lysosomal function. Recent studies have implicated MFSD12 as an oncogenic factor in several cancers, driving cell cycle progression and tumor cell proliferation, notably in melanoma and lung adenocarcinoma. Mechanistically, MFSD12 acts upstream of PI3K/AKT signaling, and its inhibition may suppress tumor growth but could also affect normal lysosomal processes. MFSD12 is increasingly considered a potential therapeutic target and prognostic biomarker, especially in oncology and lysosomal storage disease contexts.
Drugs inhibiting PI3K/AKT downstream signaling can counteract MFSD12-driven cell proliferation in melanoma. In theory, potential small-molecule inhibitors of the transporter function are being considered as research advances.
