Overview

Tumor antigen presentation via major histocompatibility complex (MHC) class I and II pathways is a fundamental process in the immune system’s recognition of cancer cells. MHC molecules display peptide fragments (antigens) on the cell surface, enabling T cells to distinguish between self and non-self, including tumor-derived antigens. The MHC class I pathway presents endogenous antigens to CD8+ T cells, leading to direct tumor cell lysis. The MHC class II pathway presents exogenous antigens to CD4+ T cells, which coordinate broader immune responses. Manipulating these pathways forms the basis for many cancer immunotherapies.
