Overview

Major histocompatibility complex class I heavy chain H-2K^b is a membrane glycoprotein expressed on nearly all nucleated cells in mice and is a prototypic mouse MHC class I alloantigen. It consists of a heavy alpha chain (~44 kDa) noncovalently associated with beta-2 microglobulin, forming a heterodimeric complex[1][4]. The peptide-binding groove of H-2K^b presents short (8–10 amino acid) peptides derived from intracellular proteins to CD8+ cytotoxic T lymphocytes, initiating antigen-specific immune responses by enabling T cell receptor recognition[1][4][5][7]. The peptide/MHC complex is stabilized by a series of hydrogen bonds at both peptide termini, and diversity in the binding groove specifies peptide compatibility and allele specificity[1][5][7]. H-2K^b is extensively used in immunological research as a model for understanding antigen processing, immune recognition, and the structural basis of T cell activation, as well as for developing murine models of infection, cancer, and immunotherapy. It is analogous in function and structure to human HLA-A alleles[1][6][7][9].
