Overview

The Major Histocompatibility Complex (MHC) class I presenting Ovalbumin 257-264 (SIINFEKL) is a cornerstone model system in immunology, primarily used to study the interaction between antigen-presenting cells and CD8+ T cells. It consists of the murine H-2Kb MHC class I molecule bound to the 8-amino acid peptide SIINFEKL, which is the immunodominant epitope of chicken egg ovalbumin (UniProt P01012; Rock et al., 1990, J Immunol). This complex is specifically recognized by the OT-I T-cell receptor, making it an essential tool for tracking antigen-specific immune responses in transgenic mouse models (Hogquist et al., 1994, Cell). Biologically, it represents the culmination of the endogenous antigen processing pathway, where intracellular proteins are processed and displayed for immune surveillance. In therapeutic research, the complex is targeted by TCR-like antibodies, such as 25-D1.16, and serves as a benchmark for evaluating the efficacy of cancer vaccines and adoptive cell therapies (Porgador et al., 1997, Immunity). While primarily an experimental target, it provides critical insights into the mechanisms of T-cell memory, exhaustion, and the development of therapies against intracellular tumor antigens.

Mechanism of action

The complex presents the SIINFEKL peptide on the cell surface for recognition by specific T-cell receptors (TCRs) on CD8+ cytotoxic T lymphocytes, initiating an immune response against cells expressing the antigen (Pamer & Cresswell, 1998, Annu Rev Immunol).
