Overview

The Major histocompatibility complex class II–glycoprotein 100 peptide complex is a molecular assembly consisting of a peptide fragment derived from the glycoprotein 100 (gp100/PMEL) protein bound within the groove of a Major Histocompatibility Complex (MHC) class II molecule (UniProt P17667). This complex is primarily expressed on the surface of professional antigen-presenting cells and, in some cases, melanoma cells themselves (PubMed: 15155838). Its primary biological role is to present the gp100 antigen to CD4+ T-helper cells, which is a critical step in orchestrating a robust and sustained anti-tumor immune response (PubMed: 9151710). In the context of oncology, this complex is a significant therapeutic target for melanoma, as gp100 is highly overexpressed in melanocytic tumors. Therapeutic strategies targeting this complex include peptide-based vaccines and adoptive T-cell therapies designed to enhance the recognition and destruction of melanoma cells (PubMed: 21632944). By engaging CD4+ T cells, these therapies can promote cytokine secretion and provide necessary help for CD8+ cytotoxic T cell activity. However, targeting this complex carries risks of off-target effects on healthy melanocytes, potentially leading to autoimmune conditions such as vitiligo or uveitis.
