Overview

The major histocompatibility complex class II–peptide antigen complex on dendritic cells is a molecular assembly in which a dendritic cell presents short, processed peptide antigens bound to an MHC class II molecule at its cell surface. This complex is essential for the activation of antigen-specific CD4^+^ helper T cells, which scan the surfaces of dendritic cells for non-self, tumor, or altered-self epitopes. Formation of the complex is initiated when dendritic cells internalize extracellular proteins, degrade them to peptides in endosomal compartments, and load them onto newly synthesized MHC class II molecules. Upon DC activation, peptide–MHC II complexes are transported to the plasma membrane, where they serve as a critical signal to naïve or memory CD4^+^ T cells, triggering a cascade of immune activation, cytokine secretion, and immune memory formation. This presentation is central for immune surveillance, vaccination efficacy, tumor immunity, and maintenance of tolerance[1][2][3][4][5][6][7].
