Overview

The Major Histocompatibility Complex class II (MHC II) presenting SARS-CoV-2 peptides on artificial antigen-presenting cells (aAPCs) is a synthetic immunotherapy platform designed to induce or expand antigen-specific CD4+ T-cell responses (NexImmune, 2021). This complex consists of MHC II molecules, such as HLA-DR, loaded with immunodominant epitopes from the SARS-CoV-2 virus, typically anchored to a nanoparticle or cellular scaffold (Oelke et al., 2003). Its primary biological function is to mimic the natural process of antigen presentation, providing the necessary signals to activate the T-cell receptor (TCR) on helper T cells (Grifoni et al., 2020). In the context of COVID-19, this system is used to bolster the immune response, particularly in individuals with weakened immunity or to provide protection against emerging viral variants (PubMed: 33097835). By delivering precise antigenic signals, these aAPCs can coordinate a comprehensive immune response involving both B cells and cytotoxic T cells. However, therapeutic use requires careful monitoring for potential safety concerns such as cytokine release syndrome or unintended immune activation (Nature, 2020).
