The major homology region (MHR) is a highly conserved stretch of approximately 20 amino acids within the capsid (CA) domain of the Gag polyprotein in all orthoretroviruses and some retrotransposons. It is essential for proper formation and maturation of the viral capsid shell, which encases the viral replication complex. The MHR plays a crucial role in viral assembly and infectivity, contributing significantly to overall capsid structure and stability. Mutations within this region disrupt membrane binding affinity, impair particle assembly, block infectivity, or result in non-infectious particles.
