Target
Der p 1, Der p 2, Der p 23
Molecular classification
Other, Allergen protein, Cysteine protease (Der p 1), Lipid-binding protein (Der p 2), Peritrophin-like protein (Der p 23)
01

Overview

Major house dust mite allergens are a group of proteins derived primarily from *Dermatophagoides pteronyssinus* that are recognized by the immune system of allergic patients, with Der p 1 (cysteine protease), Der p 2 (lipid-binding protein homologous to NPC2 and MD-2), and Der p 23 (peritrophin-like chitin-binding protein) being the most clinically important[1][5][3]. These allergens are responsible for the majority of IgE reactivity in house dust mite-allergic individuals and are major triggers for asthma, rhinitis, and atopic eczema. They are used as major molecular components in allergy diagnostics and specific immunotherapy, and they interact directly and indirectly with immune pathways, including protease activity (Der p 1), lipid/cholesterol binding (Der p 2), and innate immune recognition.

02

Mechanism of action

Induction of IgE-mediated hypersensitivity; Activation of basophils and mast cells; Upregulation of immune activation markers (e.g., CD203c by Der p 23); Some (e.g., Der p 2) can mimic or activate TLR4 pathways

03

Biological functions

Immune response induction (IgE binding and allergic sensitization)Allergenicity (triggers allergy symptoms)Protease activity (Der p 1)Lipid/cholesterol binding (Der p 2)Interaction with innate immunity pathways (Der p 2, Der p 7)
04

Disease associations

Allergy (primary driver of allergic sensitization)AsthmaAllergic rhinitisAtopic dermatitis
05

Safety considerations

High allergenic potential: may induce severe allergic symptoms, anaphylaxis in sensitized individualsVariability between individuals: not all patients are sensitized to the same combination of major allergens, complicating therapy and diagnosis
