Major outer membrane porin PorB (PorB) is a critical surface protein and voltage-gated ion channel found in pathogenic Neisseria species, including Neisseria meningitidis and Neisseria gonorrhoeae (NIH, ResearchGate). It is essential for bacterial viability, facilitating the exchange of ions and the uptake of small nutrient molecules like sugars and amino acids across the outer membrane (NIH, ASM). Beyond its metabolic functions, PorB acts as a potent virulence factor by interacting with host receptors such as TLR2 to modulate immune signaling and by binding complement regulators like Factor H and C4b-binding protein to evade host-mediated killing (NIH, Frontiers in Microbiology). In clinical settings, PorB is a major component of outer membrane vesicle (OMV)-based vaccines, where it serves as an antigen to elicit protective bactericidal antibodies (ResearchGate). However, its high degree of antigenic variation and its role in mediating antibiotic resistance—where mutations in the porin channel restrict the influx of drugs like cephalosporins and fluoroquinolones—present significant challenges for therapeutic intervention (NIH, Antimicrobial Agents and Chemotherapy). The protein exists in different allelic forms, such as PorBIA and PorBIB in gonococci, which correlate with the severity and dissemination of the infection (NIH).
