The Major outer membrane protein (MOMP) is a critical structural and functional component of the outer membrane in Gram-negative bacteria, most notably within the genus Chlamydia. It is a 40-kDa cysteine-rich protein that functions as a porin, forming beta-barrel channels that facilitate the diffusion of essential solutes and nutrients across the bacterial envelope [5, 7, 11]. Beyond its transport role, MOMP is essential for maintaining the structural rigidity of the infectious elementary body (EB) through disulfide bond cross-linking and serves as a primary adhesin for host cell attachment and entry [4, 11, 19]. In clinical medicine, MOMP is the immunodominant antigen used to define Chlamydia trachomatis serovars and is the leading candidate for vaccine development due to its surface exposure and ability to elicit neutralizing antibodies [3, 8, 18]. Therapeutic strategies targeting MOMP include recombinant subunit vaccines and novel affitoxins designed to neutralize infection or deliver lethal payloads directly to the pathogen [2, 9, 18].
