Overview

Major pollen allergen Art v 1 is a modular glycoprotein from mugwort (*Artemisia vulgaris*) pollen, consisting of a defensin-like cysteine-rich N-terminal domain and a hydroxyproline-rich, heavily O-glycosylated C-terminal region. Art v 1 elicits strong IgE-mediated immune responses in sensitized individuals and is responsible for most mugwort pollen allergy cases. Its epitopes are mainly conformational, residing on the defensin domain and transitional regions, and the protein is extremely stable due to disulfide bonds. Recombinant non-glycosylated Art v 1 is considered a promising scaffold for engineering hypoallergenic vaccines and immunotherapies. Art v 1 is also a member of a protein family present in multiple *Artemisia* species and shows considerable cross-reactivity among weed pollen allergens.

Mechanism of action

For vaccines: induction of blocking IgG antibodies and suppression of allergen-specific TH2 response. For immunotherapy: immune tolerance induction using hypoallergenic or recombinantly engineered Art v 1.
