Overview

The Major surface glycoprotein (MSG) is the most abundant surface protein of the opportunistic fungus Pneumocystis jirovecii, the causative agent of Pneumocystis pneumonia (PCP) (Stringer, 2007). It serves as a primary adhesin, mediating the attachment of the fungus to host alveolar type I pneumocytes and extracellular matrix proteins such as fibronectin and vitronectin (Kutty et al., 2013). MSG is encoded by a large family of approximately 80-100 genes, which allows the pathogen to undergo frequent antigenic variation to evade host immune surveillance (UniProt, 2024). This genetic diversity presents a significant challenge for vaccine development but also makes MSG a valuable biomarker for the diagnosis of PCP through the detection of specific antibodies or DNA sequences in clinical specimens (PubMed, 2021). While current clinical therapies like trimethoprim-sulfamethoxazole target metabolic pathways, MSG is a primary focus for the development of passive immunization strategies and prophylactic vaccines aimed at preventing infection in immunocompromised individuals (Gigliotti & Wright, 2012).
