Overview

Malate dehydrogenase (MDH) is a ubiquitous enzyme found in all domains of life, playing a central role in cellular metabolism. It catalyzes the reversible conversion of L-malate to oxaloacetate, using NAD+ (or sometimes NADP+) as a cofactor. This reaction is crucial for the citric acid cycle and other metabolic pathways. In eukaryotes, it exists as two main isoforms: Mitochondrial MDH (m-MDH) and Cytosolic MDH (c-MDH or s-MDH). MDHs are being explored as therapeutic targets, and inhibitors are under investigation as both broad-spectrum agents and isoform-selective drugs.
