Overview

MAM and LDL-receptor class A domain-containing protein 1 (MALRD1) is a conserved, protein-coding gene product containing multiple MAM and LDLR A2 domains[1][7]. Its expression is enriched in the small intestine and is upregulated during the differentiation of human intestinal epithelial cells[1][3]. MALRD1 plays a significant role in regulating the levels of fibroblast growth factors FGF15/19 in enterocytes, which are key signals in the control of bile acid metabolism in the liver[2][7]. It enhances the production and/or transport of FGF19 and thus influences the synthesis of bile acids[1][7]. Variants in MALRD1 may modulate the risk of Alzheimer's disease, particularly via synergistic interaction with APOE E4 allele[1]. Currently, there are no known drugs or biomarkers that selectively target MALRD1, and specific safety issues have not been reported in the available literature[1][2][7].
