Overview

MAM domain-containing protein 2 (MAMDC2) is a secreted, glycosylated protein characterized by four sequential MAM (meprin/A5 antigen/protein tyrosine phosphatase mu) domains and is encoded by the MAMDC2 gene[1][2][3]. This protein is present in the extracellular matrix and synthesized in the endoplasmic reticulum. MAMDC2 acts as an extracellular regulator of cell proliferation and migration, with context-dependent functions across different cancers. It serves as a tumor suppressor in ER-positive breast cancer by inhibiting the MAPK (RAF–ERK) pathway[1]. In contrast, it is upregulated in gastric cancer, promoting tumor proliferation and invasion, and is linked to poor prognosis[3]. MAMDC2 expression has also been implicated in colorectal and pancreatic cancers. Its variable function across tumor types suggests complex roles in cell signaling and tumor biology, likely through extracellular mechanisms and interaction with key regulatory pathways such as TGF-β and MAPK. The protein itself has shown promise as a biomarker for several cancers but is not currently established as a druggable therapeutic target, nor are there drugs known to specifically modulate its activity[1][3].
