Overview

Mammalian diaphanous-related formin 1 (mDia1/DRF1) is a Rho GTPase effector protein that regulates the actin and microtubule cytoskeletons. It is involved in various cellular processes such as cell migration, cytokinesis, phagocytosis, and vesicle trafficking. Activation occurs upon binding to active Rho-family GTPases, leading to actin polymerization and microtubule stabilization. HIV-1 exploits DRF functions to facilitate virus transport within host cells.

Mechanism of action

Upon activation by small GTPases, mDia1 promotes actin polymerization/nucleation at the barbed end of actin filaments and stabilizes microtubules.
