Overview

Mammalian orthoreovirus (reovirus) is a double-stranded RNA (dsRNA) virus used as an oncolytic agent to selectively infect and kill cancer cells. It preferentially replicates in transformed cells with activated Ras signaling, leading to cell death via apoptosis, necroptosis, and autophagy. Reovirus also modulates the tumor microenvironment by stimulating immune responses and reducing immunosuppression. It is under investigation as a form of cancer virotherapy, often in combination with chemotherapy, radiation, or immunotherapies.

Mechanism of action

Selective replication in cancer cells with activated Ras signaling pathways, leading to cell lysis and immunomodulation. Induces apoptosis, necroptosis, and autophagy in tumor cells. Stimulates innate and adaptive immunity and reduces MDSC suppression.
