Overview

The mammalian target of rapamycin complex 2 (mTORC2) is a multi-subunit protein kinase complex that plays a central role in regulating cell growth, survival, metabolism, and cytoskeletal organization. It consists of mTOR, Rictor, SIN1, mLST8, and variably associated components like Protor/PRR5/PRR5L and DEPTOR. mTORC2 regulates key cellular processes including cell proliferation, survival, metabolism and actin cytoskeleton organization. Aberrant activation is linked to various cancers making it an important therapeutic target.

Mechanism of action

Inhibition of mTORC2 kinase activity, although native mTORC2 is acutely insensitive to rapamycin. Prolonged exposure can disrupt assembly and affect function.
