Candida mannan is a complex, highly branched polysaccharide that constitutes a significant portion of the outer cell wall of Candida species, most notably Candida albicans. It is primarily composed of mannose residues linked via alpha-1,6, alpha-1,2, and alpha-1,3 glycosidic bonds, often featuring phosphorylation that contributes to the cell's negative surface charge (Mora-Montes et al., 2009, PubMed). This molecule serves as a critical pathogen-associated molecular pattern (PAMP) that is recognized by the host's innate immune system through various pattern recognition receptors, including Dectin-2 and the mannose receptor (Saijo et al., 2010, Nature). In clinical practice, the Candida mannan antigen is a major diagnostic target; its presence in serum is used as a biomarker for the early detection of invasive candidiasis, often preceding positive blood cultures (Mikulska et al., 2010, Critical Care). While not the primary target of current frontline antifungal drugs like azoles or echinocandins, mannan is the focus of intensive research for the development of diagnostic assays, protective vaccines, and therapeutic monoclonal antibodies aimed at enhancing fungal clearance (Sendid et al., 2015, Journal of Clinical Microbiology).
