Mannose-containing glycoproteins refer to a class of glycoproteins characterized by the presence of multiple terminal mannose residues in their N-linked carbohydrate chains. This structure arises during early glycosylation in the endoplasmic reticulum, where the glycoprotein receives an “en bloc” addition of a Glc3Man9GlcNAc2 precursor, which, after glucose removal, can retain several mannose residues (typically five to nine) on their chitobiose core. High-mannose glycoproteins are recognized by pattern recognition receptors, most notably the mannose receptor (CD206), facilitating their uptake and clearance by macrophages and liver cells. They play crucial roles in immunity, protein folding, and serve as markers of pathogens (e.g., viruses, fungi). Their exposure on cell surfaces or viral proteins can trigger innate immune responses and are involved in disease processes, including infection susceptibility, immune clearance, cancer, and congenital disorders of glycosylation.
