Overview

The Mannose receptor C type 1 (CD206) is a 180 kDa transmembrane glycoprotein and a prominent member of the C-type lectin superfamily, primarily expressed on macrophages and dendritic cells (UniProt: P22897). It functions as a pattern recognition receptor (PRR) that specifically recognizes and internalizes glycoconjugates containing terminal mannose, fucose, or N-acetylglucosamine (GlcNAc) residues, such as the N-acetylglucosamine β-mannopyranosyl residues found in complex N-glycans (PMID: 1554638). This recognition is mediated by multiple carbohydrate recognition domains (CRDs) and plays a vital role in the innate immune response by facilitating the phagocytosis of pathogens like Mycobacterium tuberculosis and Candida albicans (PMID: 11085990). Beyond its role in infection, CD206 is a well-established marker for M2-polarized macrophages, which are involved in tissue repair, inflammation resolution, and tumor progression. In the pharmaceutical industry, CD206 is targeted for diagnostic purposes, such as with the FDA-approved imaging agent Technetium Tc 99m tilmanocept, which binds the receptor to identify sentinel lymph nodes (DrugBank: DB09103). Additionally, researchers are exploring CD206-targeted delivery systems, including mannosylated liposomes and nanoparticles, to deliver therapeutic agents directly to tumor-associated macrophages or to modulate immune responses in chronic inflammatory diseases.
