MAP kinase-activated protein kinase 5 is a serine/threonine kinase encoded by the MAPKAPK5 gene in humans. It is activated by phosphorylation through upstream MAP kinases such as ERK and p38, especially in response to cellular stress and proinflammatory cytokines. The enzyme modulates signal transduction pathways by phosphorylating downstream substrates, including HSP27, influencing cellular processes including inflammation, survival, and apoptosis. It has been investigated as a therapeutic target in disorders such as rheumatoid arthritis and may play roles in neurodegenerative and cardiovascular diseases. Reduced levels of MAPKAPK5 have been linked to Alzheimer's disease, though clinical significance is yet to be fully established.
