Overview

MAPK-regulated corepressor-interacting protein 1 (MCRIP1) is a protein that acts as a molecular switch controlling epithelial-mesenchymal transition (EMT). Its function is regulated by phosphorylation: unphosphorylated MCRIP1 binds and inhibits the transcriptional corepressor CTBP(s), preventing repression of E-cadherin and thus inhibiting EMT. Upon phosphorylation by MAPK/ERK, MCRIP1 releases CTBP(s), leading to transcriptional repression of the E-cadherin gene and induction of EMT. MCRIP1 is primarily cytoplasmic but can also localize to the nucleus and cytoplasmic stress granules. While MCRIP1 has a role in cell fate transitions such as EMT—which is fundamental to cancer metastasis—direct evidence for its current use as a therapeutic target or drug interaction is lacking[1][2][3][5][9].
