Marburg virus VP24 protein (VP24)

Target
VP24
Molecular classification
Viral structural protein, Matrix protein (minor matrix protein)
01

Overview

Marburg virus VP24 protein is a minor matrix protein integral to the viral life cycle. It is involved in the assembly of the viral nucleocapsid and in budding of new virus particles from the host cell. VP24 colocalizes with nucleoprotein inclusions in infected cells and displays membrane-binding properties but cannot form virus-like particles (VLPs) independently—it is incorporated into VLPs by association with the major matrix protein VP40. Inhibition or silencing of VP24 reduces release of Marburg virions. Structurally, Marburg VP24 has a single-domain alpha/beta fold with a unique projecting β-shelf, distinct from ebolavirus VP24, and does not possess the strong immunosuppressive function of its Ebola counterpart[1][3][4][8].

Mechanism of action

Experimental antivirals could act by inhibiting VP24’s interaction with other viral proteins or with membranes, thereby impairing virion assembly and release

03

Biological functions

Assembly of the viral nucleocapsidVirion buddingInteraction with nucleoprotein and other matrix proteins
04

Disease associations

Infection (specifically, Marburg virus disease, a severe viral hemorrhagic fever)
05

Safety considerations

No direct VP24 inhibitors in clinical use mean no established safety concerns; theoretical safety issues could relate to targeting viral structural proteins in human infection
06

Interacting drugs

None currently established in clinical development; research for direct inhibitors is ongoing but no approved drugs target VP24 specifically
07

Biomarkers

VP24 protein or mRNA may be used as a viral replication or infection marker in research; no validated clinical biomarker use
