MARCKS cis regulating lncRNA promoter of cytokines and inflammation (MROCKI) is a cis-acting long non-coding RNA that is induced in human macrophages by Toll-like receptor (TLR) activation. MROCKI regulates the expression of its neighboring gene, MARCKS (myristoylated alanine-rich protein kinase C substrate), by forming a ribonucleoprotein complex with APEX1 at the MARCKS promoter. This complex recruits histone deacetylase HDAC1, leading to changes in chromatin acetylation and suppression of MARCKS transcription. The downstream effects include enhanced production of inflammatory cytokines and chemokines. Genetic variants that influence MROCKI expression are associated with altered risk for several inflammation-related and infection-related diseases, including inflammatory bowel disease and tuberculosis. The lncRNA functions as a master regulator of macrophage innate responses, making it a molecule of interest in studying immune regulation and inflammatory diseases[1][6][8].
