MARVEL domain-containing protein 3 (MARVELD3) is a member of the tight junction–associated MARVEL protein (TAMP) family, which includes occludin and tricellulin[1]. It is a transmembrane protein containing a conserved MARVEL domain, characterized by four transmembrane helices[1][4]. MARVELD3 localizes to epithelial tight junctions, where it contributes to the regulation of paracellular ion permeability and epithelial barrier function[1][2][5]. Although MARVELD3 can partially compensate for the loss of other TAMPs, it has distinct, nonredundant roles in tight junction assembly and maintenance[1]. MARVELD3 is expressed across multiple epithelial tissues and its abnormal expression has been implicated as a potential prognostic marker in various cancers, including oral squamous cell carcinoma and hepatocellular carcinoma[2]. There are currently no approved drugs directly targeting MARVELD3, and no known safety liabilities associated with modulating this protein.
