The Mas-related G protein-coupled receptor member B3 (MrgprB3 or MrgB3) is an orphan receptor that belongs to the Mrgpr family, predominantly found in rat mast cells and subsets of sensory neurons. It plays a role in mediating mast cell degranulation in response to certain peptides, notably pituitary adenylate cyclase-activating peptides such as PACAP-38 and its analogs. MrgprB3 is functionally analogous to the human MRGPRX2 receptor, which is involved in non-IgE-mediated mast cell activation and neurogenic inflammation. The biological relevance of MrgprB3 primarily relates to the transduction of pain and itch signals and the regulation of mast cell-mediated inflammatory responses, as suggested by in vitro and animal studies.
Ligand-induced activation of the MrgprB3 receptor on mast cells leads to mast cell degranulation and the release of pro-inflammatory mediators, contributing to neurogenic inflammation.
