Mas-related G-protein coupled receptor member E (MRGPRE) is an orphan member of the Class A (Rhodopsin-like) GPCR superfamily, primarily predicted to function in the modulation and/or development of nociceptors, including roles in the sensation or modulation of pain. It is mainly expressed in sensory neurons, especially in the peripheral nervous system. Unlike some other MRGPR family members with established ligands, MRGPRE does not have a well-characterized endogenous agonist or antagonist; its precise signaling properties and physiological ligands remain undefined. Structurally, MRGPRE shares unique features seen in MRGPRs—including a shallow, solvent-exposed orthosteric pocket and absence of some canonical GPCR motifs—which drive ligand specificity and potentially rapid evolutionary adaptation. Research suggests it may play a role in neuronal communication relevant to pain and itch, but its clinical relevance as a direct therapeutic target is not fully established[1][3][4][5][7][9][10].
