MAS1 proto-oncogene, G protein-coupled receptor, is a transmembrane class I GPCR that primarily functions as the receptor for angiotensin-(1-7), a peptide hormone of the renin-angiotensin system. MAS1 modulates multiple signaling pathways—most notably, it antagonizes the effects of angiotensin II via the type 1 receptor (AGTR1), thus playing protective roles in the cardiovascular system such as lowering blood pressure and promoting vasodilation. In addition, MAS1 activation regulates cell growth and metabolic functions, showing anti-proliferative, anti-migratory effects in cancer models and controlling adipogenesis and thermogenesis in metabolic tissues. It is expressed in a wide range of tissues, particularly those involved in cardiovascular, metabolic, reproductive, and neurological functions. Aberrant MAS1 signaling has implications in diseases such as hypertension, cancer, metabolic disorders, and reproductive pathologies. Research interest persists in targeting this receptor for drug development due to its beneficial roles in these conditions and its position as a nodal modulator in cell signaling.
Activation via angiotensin-(1-7) leads to Gq protein coupling, activation of phospholipase C, calcium entry modulation, downstream MAPK and NF-κB signaling, PI3K/Akt pathway activation. Antagonism of AGTR1 signaling, reducing detrimental actions of angiotensin II in cardiovascular and metabolic tissues.
