Mast cells are tissue-resident immune cells crucial in allergic reactions, inflammation, and host defense. They release mediators like histamine, heparin, and cytokines upon activation via various mechanisms, including IgE cross-linking, DAMP/PAMP recognition, and complement activation. Dysregulation leads to diseases like asthma, mastocytosis, and MCAS.
Drugs targeting mast cells aim to inhibit their activation, mediator release, or downstream effects. Antihistamines block histamine receptors, mast cell stabilizers prevent degranulation, epinephrine reverses anaphylaxis, anti-IgE antibodies reduce IgE levels, and corticosteroids suppress inflammation.
