Maternal embryonic leucine zipper kinase (MELK) is a serine/threonine protein kinase involved in cell cycle regulation, stem cell self-renewal, apoptosis, splicing regulation, and gene expression. It plays a role in embryonic development, hematopoiesis, and oncogenesis, with high expression observed in various cancers. Pharmacological inhibition of MELK has shown preclinical efficacy in impairing tumor growth, but recent studies suggest its necessity may be context-dependent. Clinical trials are ongoing for inhibitors targeting MELK.
