Overview

MATN1 antisense RNA 1 (MATN1-AS1) is a long non-coding RNA (lncRNA) transcribed from the antisense strand of the MATN1 locus. It does not code for a protein. Research shows MATN1-AS1 plays important regulatory roles in multiple cancers, functioning through post-transcriptional and transcriptional mechanisms. In glioblastoma and glioma, MATN1-AS1 can act as a tumor suppressor or oncogene, depending on cellular context. In glioblastoma, its upregulation inhibits cell proliferation and invasion and promotes apoptosis by downregulating RELA via E2F6 and suppressing the MAPK signaling pathway[1]. In glioma, MATN1-AS1 promotes tumor progression by sponging microRNAs (miR-200b/c/429), leading to upregulation of the oncogene CHD1[4]. MATN1-AS1 also plays roles in colorectal cancer via a similar ceRNA network (regulating miR-200c-3p and HAS2)[2] and in osteosarcoma prognosis[3]. Given these pleiotropic effects, MATN1-AS1 is considered a potential therapeutic target and biomarker in several cancers, though it is not a classic receptor, enzyme, or transporter. No direct small-molecule drugs or therapies currently target MATN1-AS1, but its regulatory networks may provide intervention points.

Mechanism of action

Regulates gene expression by acting as a ceRNA (competing endogenous RNA) for specific microRNAs (e.g., miR-200b/c/429, miR-200c-3p), binds transcription factors (e.g., E2F6) affecting target gene transcription (e.g., RELA, CHD1), modulates signaling pathways (MAPK pathway)
