Matrilin-1 is a cartilage-specific extracellular matrix protein encoded by the MATN1 gene. It is a member of the von Willebrand factor A domain containing protein family and is one of the major non-collagenous structural proteins in cartilage. Matrilin-1 forms homotrimeric and hetero-oligomeric fibrils (with matrilin-3), binds to type II and IX collagens and aggrecan, and is required for the proper network-like organization and bundling of collagen fibrils in the cartilage extracellular matrix. It is primarily expressed in cartilage by chondrocytes in the maturation zone of the growth plate. Mutations in MATN1 have been associated with inherited chondrodysplasias and idiopathic scoliosis. Although matrilin-1 plays a crucial structural role in cartilage, there is no evidence of direct targeting by drugs or therapeutic interventions
