The Matrix 2 protein (M2) is a small, integral membrane protein found in the viral envelope of influenza A virus. It functions as a proton-selective ion channel (viroporin), playing essential roles in the virus life cycle, particularly during entry, assembly, and budding. M2 conducts protons across the viral membrane, facilitating uncoating by loosening ribonucleoprotein complexes from M1. The N-terminal extracellular region (M2e) is a highly conserved target for universal vaccine development. Resistance to M2 inhibitors like amantadine and rimantadine is a significant therapeutic challenge.
